The manual is published initially as a core text of basic techniques, to which will be added supplementary chapters and sections at regular intervals. One characteristic of plant tissue culture methodology is the fact that what works for one species quite possibly will not work for another (or even for a different genotype of the same species): each requires a carefully optimized protocol, whether it be for callus culture initiation, protoplast isolation and culture or plant regeneration following Agrobacterium-mediated transformation. With this in mind, we have decided on the one hand to generalize experimental approaches for some basic techniques, by reference to the use of model species. This should at least give the researcher a feel for the techniques, and indeed provide a starting point for species-specific optimization of protocols. On the other hand, we have provided detailed protocols for the manipulation of a number of specific crop species, which have been difficult to optimize and for which there may be particular interest. A detailed bibliography is provided in each chapter, so that access to the relevant literature is possible.
